Week 2- Human Cells and the Basics of Using a Microscope
The Microscope;
-Components
-The powers of ten (letter e)

Types of slide
-whole mount
This type of slide involves fixing an entire specimen to a slide beneath a coverslip. This may result in the slide being thick.
-smear or drop
-squash
-section

Examine the following slides and identify the structures in each;
•Anterior lobe of pituitary gland (ls) stained with hematoxylin and eosin (H&E).
-acidophils (cytoplasm stained bright red/pink)
-Basophils (stained deep blue/purple)
-Chromophobes (absence of stain causes grey color; clear nucleus with pale cytoplasm and
indistinct cell boundaries)

•Small intestine cross-section (cs) stained with sudan red and hematoxylin.
-goblet cells
-column shaped nucleated epithelial cells on the surface layer

•Simple squamous mesothelium (wm) stained with a silver stain (silver nitrate)
silver nitrate impregnation brings the epithelial borders into sharp relief.

•Sperm (smear) stained with hematoxylin. 100X objective (multiplied by 10X occular lens =
1000X total magnification).

•Adipose tissue (section) stained with hematoxylin and sudan red.

•Embrionic Germ Layers of a Gastrula (pluripotent stem cell)
-Ectoderm (outside)
-Mesoderm (middle)
-Endodern (inside)
The Microscope;


The lowercase letter "e" is used here to demonstrate the range of the powers of ten. Occular lenses commonly have a magnification of ten times (10X). This number is multiplied by the magnifying power of the objective lense (typically 4X, 10X, 45X, or 100X) to find
the overall magnification.
